We propose to develop a method and an instrument aiming at surpassing current technologies of DNA sequencing, in terms of rapidity and reliability. The method relies on the development and the use of mutant polymerase with refined thermodynamical properties. The instrument will be based on nanostructures allowing the fluorescence detection of nucleotide capture events by these unique polymerases. These nanostructures will be designed to allow a temporal control of the temperature at the nanoscale in order to control polymerisation. Within the course of the project, the process will illustrating by sequencing reference DNA (M13mp18) as a means to evidence a proof of concept
