FP-BioPrev

Postoperative mortality of pancreatectomy is 5% and morbidity of this surgery is high with a complication rate around 50%. One of the most serious complications of pancreatectomy is the occurrence of pancreatic fistula (PF). A FP is a wound healing and sealing of intestinal anastomosis and leakage of pancreatic enzymes into the peritoneal environment. These enzymes digest vessels and organs near the pancreas and thus may become life threatening. To date, no biomaterial available on the market is indicated for the prevention of the onset of FP following pancreatectomy. Thus, this project aims to design and validate a biomaterial dedicated to the prevention of the occurrence of FP. This project aim to develop and validate a biomaterial in the form of a double film layer: (1) a biopolymers matrix in contact with pancreas that will provide the dual function of sponge (retaining the pancreatic enzymes released if fistula) and antibacterial agent reservoir (to prevent the risk of infection), and (2) a backing layer impermeable to pancreatic enzymes to limit their dissemination in the peritoneal environment. The first layer must not be only bioadhesive but should also be able to exhibit special adhesive properties to the organ according to the surrounding temperature. The originality of the FP- BioPrev project is the use of biocompatible natural polymers for the preparation of the biopolymers matrix followed by surface functionalization to obtain the desired surface characteristics. This surface modification would provide controlled adhesive properties, which will facilitate the positioning of the biomaterial. The idea is to get an easily handled and repositionable device by the surgeon during the procedure (at room temperature) which, once positioned and warmed to body temperature present enhanced bioadhesive properties. Thus, a new "smart" medical device, containing a matrix with conventional properties of biopolymers, but with advanced surface properties will be developed. The objectives of this project are within the context of societal challenge "Health and Wellbeing " and especially under Axis 4 " Biomedical Innovation ." Indeed, the development of this biomaterial may enable the reduction of postoperative complications and thus reduce hospitalization costs, which is part of the optimization of health policy context. To complete this project the chosen instrument, "Young Researchers”, aims to prepare young researchers talents to become future leaders of the French scientific research . Indeed, thanks to this research project the researcher can build skills project management, coaching and gain solid experience in managing a project and strengthening collaborations recently established in France and abroad.
