Immune checkpoint inhibitor (ICI) therapy has revolutionized the cancer field. By (re)activating the immune system, even advanced stages of various types of cancer have become treatable. However, a large group of cancer patients do not respond well to ICI therapy due to unknown reasons. The bacterial microbiota in the gut has been implicated to influence ICI therapy response via crosstalk with the immune system. In this proposal, we will elucidate the role of the understudied immune-activating bacterial product ADP-heptose in ICI therapy response. Our ultimate goal is to improve the outcome of ICI therapy in cancer patients.
